A study on the activities of praziquantel Distocide^(¢ç) (against larvae) and adult worms of Clonorchis sinensis experimentally infected in rats

Abstract
Praziquantel is proven to be a highly effective therapeutic agent against all kinds of trematode and cestode infections and the drug of choice for Clonorchis sinensis(Rim, 1979).
Recently this compound was produced in Korea using a newly developed technique which enabled an easier and a less expensive manufacturing of this product. This study was conducted to observe the experimental chemotherapeutic effect of this newly developed agent, Distocide¢ç, against Clonorchis sinensis.
One metacercariae of Clonorchis sinensis were divided each into 28 groups according to dosage and developmental stage of the worm.
Thirty eight rats were treated with 1X 300 mg/kg X 1 day, 1 X 600 mg/kg X 1 day and 3 x 100x mg/kg X 2 days for the purpose of observing chemoprophylactic activity of Distocide¢ç.
Another 72 rats were treated with 3 x 100 mg/kg x 1 day and 3 x 100 mg/kg x 2 days to assess the efficacy in relation to the developmental stage of the worm and 40 rats were used as control.
Experimental animals including the controls were necropsied at 4 weeks after the medication in order to evaluate the curative activity of the drug based on the recovery rate of the worms. The number of Clonorchis eggs per gram of feces(EPG) were examined by Stoill¢¥s egg diluton counting technique once a week starting from 1 week before treatment to 4weeks after treatment.
The dosage of 1 x 30 mg/kg x 1 day of praziquantel showed a high larvicidal effect except in the cases in which medications were given at 24 hours before infection. The dosage of 3: x 100mg/kg x 2 days also showed similar effects. A complete chemoprophylactic and larvicidal effect was seen in cases given the dosage of 1 x 600 mg/kg x 1 day, recovery rates were relatively low in the early stage of Clonorchis infection as compared to those of the controls but the recovery rate increased gradually up to 10.8 % against 8 weeks old, Clonorchis sinensi s.- In the dosage of 3 x 100 mg/kg x 2 days, however only one living worm and 1 to 9 dead worms were recovered. The mean EPG at two weeks after treatment was markedly reduced and the low values of EPG were still generally noted at four weeks after treatment in both dosage groups.
According to Rim(1980, 1981), in Biltricide¢ç-a praziquantel of German made no living worms were recovered in any of the developmental stages of Clonorchis and no more Clonorchis eggs were detected at 4, weeks after medication. Above results suggest that complete elimination of worms were slightly delayed in the administration of Distocide¢ç, but, the ultimate efficacy of Distocide¢ç presented no apparent differences, thus, rendering this product as highly effective against Clonorchis sinenis.
